A question about Time and In-game time

I'm new to speedrunning and don't know much, but i couldn't find an answer to this question: If In-game time means time that shows at the end of the run (on the end screen), and normal time means when i stop a real life timer, then when do i need to stop it? Immediately after killing Dragun? |

In 1 character runs, we don't use real time, only in-game time. On multi-character runs however, we use real time only and apparently you're supposed to stop it when you shoot yourself with the gun that can kill the past in your last character's run. I might be wrong though. |
