I believe it does not, I have not finished mario challenge normally, but I am pretty sure 19 is post game, and you don't need to do it. I might be wrong.

Announcement Regarding Twitch Highlight Deletion.
Twitch will be permanently deleting user highlights for every user on the platform if your total highlights exceed 100 hours on April 19th (~2.5 weeks from making this post). Update, this has been extended to May 19th.
