The four of plateau shrines cannot be completed without getting the rune since the monk will ask you to get it. So no it is not allowed
If it was possible to do a run with it, it would be allowed. So if you somehow managed to prove us wrong and unlock the Paraglider without ever getting the rune, you could do that. Note that if you want to use the reloading of save files, all loaded files have to be from fresh playthroughs that were started during the run.
