Just try everything.^^ Either like in Jizzy(cs mission), or just holding until you sit in the car.
You can try taking it over during Los Desperados. I have never tested that.
Feels fine to me, using both the bike and a - no clue how those things are called :D the cover nearby - as cover. You can avoid the despawn like at the start of Doberman/Outrider.
You get off the bike, wait for the call, cancel it, kill him, sit through the cutscene.
I cannot remember getting any spawns ever in months of running. The valets move within a very small time-frame, there is no time for customers to spawn, let alone block the "parking zone".

Edit: Nonsense, no customers spawn because I block the "parking zone" with my car. That does not prevent the D.A. from re-spawning - it never has for me, anyways. Customer cars existing does prevent it (no matter if they get stuck in the parking zone or elsewhere).
I have forgotten what turns customers into "non-customers" - killing should.
My success rate is only 5 out of 7 so far. Since I use a teleporter, I always park in the same spot: the center of the spawn location. My angle is also always the same - I don't know what it is, anymore.
Can I have a bunch of SCRLogs, please? The log file is cleared every time you restart the game. Maybe it is necessary but not sufficient to skip it on the second frame for you - or even in general. http://www.speedrun.com/post/8qax4
I had him despawn right in front of my eyes, though. ^^ I never killed him earlier than necessary, since. But actually, I am not certain if that was before 0 star strats... Back then I used a Catalina call (the main purpose was not skipping High Noon but duping Misappropriation, Flight School and Black Project with one om0 setup).
Anyways, you had one person vanish 2s or so after killing him, so you really should collect the dossier asap. :D

Misappropriation: The only path to "dossier destroyed in the explosion" is that the guy who picked up the dossier despawns.¤
You killed two people in pretty much the same spot. Afterwards, there is only one corpse there. My untested guess is that the game just de-spawned the second guard with the dossier since there was already a corpse lying around there.
After the first mission pass, the guards like to despawn. Probably they are treated like normal peds? So keep them in your view, don't go too far away.
If you don't aim at the guard with the dossier, he will not shoot you, so I'd kill everyone except him, then kill him standing right next to him.

Jizzy: You took the call in the pre-mission cutscene (the one where CJ walks towards the building, then it fades, then the intro cs starts). That made CJ stop, so he stayed inside the marker.

I use a heuristic in my runs: when the valet in the ZR waits for the red light and then gets to the garage quickly, I block one spot, if he takes some time or I am slow, I block both.
First, you skipped on the first frame. It only starts twice if you skip later than that. After that, CJ left the marker. Finally, you skipped too late, the second instance started too early.
Afaik, it is frame perfect, you need to skip on the second frame of the cutscene. That's why it confuses me that anyone could get a 100% success rate - or does it sometimes only start once for you as well? A bunch of scrlogs would be nice, reset.
Skip the cutscene before she dies. I forgot to emphasize the importance of this in my post. :(
The final CTB cs cancels calls. You can get another call in the "wait 400" after the cutscene, and hold that. That seems to be inconsistent for Mh - maybe that is because he did not have any high priority calls in the failed attempts (it is complicated to explain^^) - or I made a mistake or things are complicated.
This should be faster.
Should be slower than the proper call holding route, though.
What's the timing on that?
If you like pain you can try hitting the lamp post to get over the bannister inside the car.

You can avoid the despawn area at the start of the mission. It is only as big as the one in Trucking, IIRC. After 3 minutes of not being inside it, the vehicle despawns - that is due to the exe, independent of missions. I did not read the exe code since I don't know how to do that. There is a slim chance that there is some way to keep the vehicle even after 3 minutes besides entering it again. If there is, you cannot get into two monster trucks, since the game only "remembers" your last two vehicles. In SA, unlike VC/III, mission vehicles can "push" vehicles out of these two slots.
