There are a few main ways to save time in an Enter the Heist speedrun.

The blind walk: As soon as the game loads, you're able to control your character without having started the timer. With this, you can press the button next to the door in the starting room before your screen even starts glitching for the first time. This takes a lot of practice at first, and can definitely be frustrating. The main semi-consistent strategy is to hold right and down without touching the mouse. After doing this for a bit, you can look slightly down and turn around, spam left click and hope.

The phone thing: In the next room, you can remove the necessity for fast reactions by clicking on the phone at specified voice lines. For the first ring, pick the phone up when the narrator is saying "ringing." it's hard to get it consistent, but at some point you'll get it consistently. After picking it up for the first time, just spam left click on it and you should pick it up pretty quickly, and you can get it without even having it ring at all if you're lucky. For the second ring, just do the same thing you did before, but now do it when the narrator is saying "things."

The table: In the next room (the white room with the curtains and shit) you can climb the stairs, drop quickly on the table, then drop to the floor. This just skips a bunch of dialogue, and you can just head through the door.
