For some reason after I input the code into the panel in the prison, the prison critters don't spawn. I've restarted my run multiple times and they still haven't spawned.

You are probably picking up the hand to late, the trigger for their spawning doesn't cover the whole survival section

You have to pick up the red hand, not just let your hand sit on it, the trigger is at the door of the red hand room and extends to about halfway through the second prison cell
