Assuming you're talking about actually having full analog input, not just mapping d-pad to analog, the only way is with the toxinput mod which is not allowed in any runs. Almost all runners of this game just use d-pad since it allows for more precision, and in my incredibly unbiased opinion the game is fun regardless

Twilightmenu is allowed, just submit it under whatever console type the twilightmenu is on

Speedrun.com for this game is really more about the actual main game, I know there are various leaderboards for the minigames in other places though

The glitch needed to beat bowser in the sky before the other two is really really hard and luck dependent and that would be very similar to any% but as the old saying goes if you want the category to be added run it yourself

"All bosses" is almost the exact same thing as all characters, an already existing category that almost no one runs, since you have to beat all of the character bosses for that. "All keys" would also be very very similar and wouldn't really have any point to it since the rabbits add very little to the actual gameplay of the run so it would just be all characters but with a bunch of extra castle movement and also 50 stars to get Wario's upstairs rabbits

That route and the strats in that run are incredibly outdated and slow, you should really just keep practicing moat skip and you'll get better at it eventually, or try to base a no moat skip route off of a more recent run

Usually with things like challenge categories there are a lot more people (cough cough Nathaniel Bandy viewers) who want to see things like "no Mario" being added to the boards but there are very rarely actual runners of the game interested in running them. Though I guess if you thought of a good enough idea there's probably someone who'd run it.
