I think 100 colonists is good. The difference between 50 and 100 is really small from a strategy perspective imo. I wouldn't add a 50.

I think all science is a bit demanding and unappealing.

A middle ground maybe First banner upgrade

Without a doubt rule change to as soon as you gain control of your character is when the timer starts. That's a general speedrun rule. I understand the desire to get a good placement but you can easily restart if in a bad spot.
