I would say yes to monkey lab (because a lot of people aren't able to disable them) and yes to specialist buildings to allow for different strategies to be viable. I don't mind special agents as they're almost required for impoppable on expert maps if you're not using double cash or injections. Allow autoplay because it's faster not to use it lol.
I would prefer without injections and double cash (and other cheats enabled). I don't mind Monkey lab or specialty buildings, but I'm happy to stop using them if the general consensus is against them.

The idea is that it splits every 10 rounds including 65 and 85, starts itself on round 1 and resets automatically. What happens when you try to use it?

I feel that for ease of autosplitting the run should end as soon as the final bloon has been popped. There's a variable that tracks whether a menu is open, closed or on a confirm screen, so you can set it to end the run on round 50 when a menu opens (congratulations screen), and reset on a confirmation screen (home button or restart level).
I watched rubixcubix to see whether to split at the end of the round or once the freeplay button was showing, which is why I did it. It would be nice to get some clarification before I try anymore runs only to find out that they could be beaten by 2 seconds due to the rules changing in the future.
Where can I find the rules tab? All I can see are rule suggestions in the forums.
I'm very new to this, but when I link the file as a "scriptable auto splitter" the timer won't start, even if I try to start it manually.
