When you're on a ladder, if you hold the direction away from the wall the ladder is on then spam the backflip button, you will be rocketed upwards with the momentum you had gained on the ladder. there are several places in the run this can be used, many of them taking place inside of the ghostly forest area, a few more in the dead forest area, and a few more in the overworld area.
