This applies to both any% and NG+, but once you defeat the yellow symbiote boss in Downtown, you'll have a "talk" with Black Cat. If you choose to help Nightcrawler (which is recommended as you'll see soon), make your way to the Residential entrance of the Sewers where he waits at the save room. Normally you'll have to help him rescue civilians from various places. However, if you enter the room enough where the symbiote forms onto the door, but the door has not closed yet (thus not locking it), and you dodge your way out of the room, the game registers this as you having completed the side task (make sure to leave though in Residential as soon as his posture changes/is about to talk). This saves a significant amount of time for runs and would have you approach the hive sooner. Note: this technique does not work for the symbiote portal room located in Pier.
