So I’m going to attempt a skipless run but do I need to be in speed run mode if so what else do I need to have on or off

If you want to play through the regular game mode there is a misc. category called full run you can submit to but it's pretty much an obsolete category. Speedrun mode will skip all the cutscenes and cut the time in half.
