Using 3 rogues and a priest would make this run faster, since we would get more crits. Using more buff plants is a great (if a little bit pay-to win) too. One player uses staff of thunder and trekking pole (and peeking in rooms) to get in the right room, and the others use the SoT and the one-punch to defeat the bosses. Another player setup is that one of the rogue's uses the elf instead to res teammates while they can't move and can't get hurt. (Our elf still didn't max out his character.) This is (imo) the optimal strat for 4-player.
